Thesis Abstract

Abstract
This research project focused on the isolation and identification of Escherichia coli as an indicator of fecal pollution in streams on the Obafemi Awolowo University (O.A.U) campus in Ile-Ife, Osun State. The presence of Escherichia coli in water bodies is a significant health concern as it is commonly found in the intestines of warm-blooded animals, including humans, and is used as an indicator of fecal contamination. The study aimed to assess the level of fecal pollution in streams within the university campus by isolating and identifying Escherichia coli strains. Water samples were collected from different streams on the O.A.U campus and analyzed for the presence of Escherichia coli using standard microbiological techniques. The samples were first subjected to membrane filtration to concentrate the bacteria, followed by culturing on selective media such as MacConkey agar and Eosin Methylene Blue agar to isolate Escherichia coli colonies. Biochemical tests, including indole production, methyl red test, Voges-Proskauer test, and citrate utilization test, were performed to confirm the identity of the isolated bacteria. The results of the study revealed the presence of Escherichia coli in the water samples collected from the streams on the O.A.U campus. The identification of Escherichia coli was confirmed based on the biochemical characteristics of the isolates. The detection of Escherichia coli in the streams indicates fecal contamination and potential health risks associated with waterborne diseases. The findings of this research highlight the importance of monitoring water quality in streams on university campuses, especially in areas where human activities can contribute to fecal pollution. The presence of Escherichia coli serves as a warning sign of potential health hazards and the need for proper wastewater management practices to prevent contamination of water sources. Regular monitoring and assessment of water quality parameters, including the presence of indicator bacteria like Escherichia coli, are essential for ensuring the safety of water resources and protecting public health. Overall, this study provides valuable information on the level of fecal pollution in streams on the O.A.U campus and underscores the importance of implementing measures to mitigate contamination and safeguard water quality for the well-being of the university community and surrounding environment.
