Thesis Abstract

Abstract
Gamma-aminobutyric acid (GABA) is the main inhibitory neurotransmitter in the central nervous system, playing a crucial role in regulating neuronal activity. GABA acts by binding to specific receptors, leading to hyperpolarization of the neuron and thus reducing the likelihood of an action potential being generated. Dysfunction in the GABAergic system has been implicated in various neurological and psychiatric disorders, such as epilepsy, anxiety, and depression. Research on GABA has shown its involvement in modulating stress responses, fear extinction, and cognitive functions. The GABAergic system interacts with other neurotransmitter systems, such as glutamate and dopamine, to maintain the delicate balance between excitation and inhibition in the brain. Imbalances in this system have been linked to the pathophysiology of several mental health conditions. Pharmacological agents that target the GABAergic system, such as benzodiazepines and barbiturates, are commonly used as anxiolytics, sedatives, and anticonvulsants. These drugs enhance GABAergic transmission, leading to a reduction in neuronal excitability. However, their clinical use is often associated with side effects, tolerance, and dependence. Recent research has focused on developing novel therapeutic strategies that target the GABAergic system more selectively and with fewer adverse effects. This includes the exploration of GABA receptor modulators, GABA reuptake inhibitors, and compounds that enhance GABA synthesis. Additionally, non-pharmacological approaches, such as neurofeedback and transcranial magnetic stimulation, have shown promise in modulating GABAergic activity in a more targeted manner. Understanding the intricate mechanisms underlying GABAergic signaling is crucial for the development of effective treatments for neurological and psychiatric disorders. Advances in neuroimaging techniques, such as magnetic resonance spectroscopy, allow for the in vivo measurement of GABA levels in the human brain, providing valuable insights into GABAergic dysfunction in various conditions. In conclusion, GABA plays a central role in maintaining the balance between excitation and inhibition in the brain, with implications for a wide range of neurological and psychiatric disorders. Further research into the GABAergic system and the development of novel therapeutic interventions hold promise for improving treatment outcomes and enhancing our understanding of brain function.
